R3 classic running like a twin. Backfiring and stumbling

I think that i am thinking in another direction.
The first thing I would do is spray starting fluid into the dead cyl while if it reacts then you have a fuel problem and if it doesn't then you have an ignition problem
On ignition problem you can use a 12 volt test light by hooking clamp to positive terminal and probe the outside terminal (the one the ecu grounds) some test lights will kill the coil and some will blink when the ecu fires.

You guys are awesome. And now I feel kinda dumb. But now I know more about my little beast so it was all worth it. The intake manifold clamp had come loose and was letting air into the first cylinder. After removing, re-seating, and tightening the clamp….. SHE Purrrrs again!!!!! Easy fix and now I have a compression tester in my toolbox and fresh spark plugs. She should be good to go for the foreseeable future.
