Pretty much. You need to look at the upper end. Also feel free to regrease the splines on the bottom end.

You mentioned you haven't owned a shaft drive before, and that spinning the wheel by hand is smooth.
Sound to me that you have back lash in the drive train, which is normal wear and tear if its within factory tolerance.
The drive shaft spline clearance and diff clearance all add up to clunking if the engine isn't pulling a load in one direction. (which is whats happening by driving the wheel in mid air)
You could try shifting into gear (engine off) whith wheel above ground and rock the wheel back and forth to listen for the noise, that's the back lash.

Having replaced two final drives and a drive shaft on my previous '11 Roadster I would suggest starting with the advice @atomsplitter gave you by removing the final drive. You almost surely need to lube the splines on that end of the driveshaft anyway. You can inspect the joint at that end of the system. Also, if you see final drive fluid there you have a leaky seal in the FD- that's what happened to me twice, which is pretty rare. If that's dry clean up and lube the splines. Hopefully, it's just normal play in the system.

Thanks, I've had the bevel box off and lubed all splines. The wheel ones too. No leaking oil, and the splines look good. I'll take the drive shaft out if the sound gets worse.
